Peptidyl-Prolyl Isomerase 1 Regulates Ca Handling by Modulating Sarco(Endo)Plasmic Reticulum Calcium ATPase and Na/Ca Exchanger 1 Protein Levels and Function

چکیده
Background-—Aberrant Ca handling is a prominent feature of heart failure. Elucidation of the molecular mechanisms responsible for aberrant Ca handling is essential for the development of strategies to blunt pathological changes in calcium dynamics. The peptidyl-prolyl cis-trans isomerase peptidyl-prolyl isomerase 1 (Pin1) is a critical mediator of myocardial hypertrophy development and cardiac progenitor cell cycle. However, the influence of Pin1 on calcium cycling regulation has not been explored. On the basis of these findings, the aim of this study is to define Pin1 as a novel modulator of Ca handling, with implications for improving myocardial contractility and potential for ameliorating development of heart failure.

متن کاملCalcium Adaptation to Na-pump Inhibition in a Human Megakaryocytic Cell Line Running Head: The Na-Ca link in megakaryocytes
The unique characteristics of the platelet Na/Ca exchanger, i.e., its dependency on both the trans-membrane Na and K gradients, render it highly sensitive to Napump inhibition. In this project we observed that the human megakaryocytic cell line CHRF-288 expresses both the α1 and α3 isoforms of the Na-K-ATPase. Inhibition of the Na-pump increased the RNA and protein expressions of the sarco(endo...
متن کاملRegulation of cardiac myocyte contractility by phospholemman: Na+/Ca2+ exchange versus Na+ -K+ -ATPase.
Phospholemman (PLM) regulates cardiac Na(+)/Ca(2+) exchanger (NCX1) and Na(+)-K(+)-ATPase in cardiac myocytes. PLM, when phosphorylated at Ser(68), disinhibits Na(+)-K(+)-ATPase but inhibits NCX1. PLM regulates cardiac contractility by modulating Na(+)-K(+)-ATPase and/or NCX1. In this study, we first demonstrated that adult mouse cardiac myocytes cultured for 48 h had normal surface membrane ar...
متن کاملRegulation of presynaptic calcium in a mammalian synaptic terminal.
Ca(2+) signaling in synaptic terminals plays a critical role in neurotransmitter release and short-term synaptic plasticity. In the present study, we examined the role of synaptic Ca(2+) handling mechanisms in the synaptic terminals of mammalian rod bipolar cells, neurons that play a pivotal role in the high-sensitivity vision pathway. We found that mitochondria sequester Ca(2+) under condition...
متن کاملModulation of sarcoplasmic reticulum function by PST2744 [istaroxime; (E,Z)-3-((2-aminoethoxy)imino) androstane-6,17-dione hydrochloride)] in a pressure-overload heart failure model.
PST2744 [Istaroxime; (E,Z)-3-((2-aminoethoxy)imino) androstane-6,17-dione hydrochloride)] is a novel inotropic agent that enhances sarco(endo)plasmic reticulum Ca(2+) ATPase (SERCA) 2 activity. We investigated the istaroxime effect on Ca(2+) handling abnormalities in myocardial hypertrophy/failure (HF). Guinea pig myocytes were studied 12 weeks after aortic banding (AoB) and compared with those...
